Abstrak
ABSTRAK Penambahan susu skim dalam bahan pengencer dapat mempertahankan kualitas spematozoa serta dapat melindungi spermatozoa selama penyimpanan. Penelitian ini bertujuan untuk mengetahui pengaruh level susu skim dalam pengencer NaCl kuning telur terhadap kualitas semen sapi Donggala selama penyimpanan pada suhu 5oC. Penelitian ini telah dilaksanakan di Laboratorium Unit Pelaksanaan Teknis Dinas (UPTD) Pembibitan Ternak, Provinsi Sulawesi Tengah, pada bulan Januari-Maret 2022. Semen berasal dari 1 ekor sapi Donggala jantan dengan kondisi reproduksi yang baik, berumur 3 tahun dengan berat badan 300 kg. Design penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan: P0 = 95% NaCl fisiologis + 5% kuning telur; P1 = 88% NaCl fisiologis + 5% kuning telur + 7% susu skim; P2 = 87% NaCl fisiologis + 5% kuning telur + 8% susu skim; P3 = 86% NaCl fisiologis + 5% kuning telur + 9% susu skim. Setiap perlakuan diulang sebanyak 5 kali. Variabel yang diamati yaitu motilitas, viabilitas, dan abnormalitas. Analisis data dengan analisis varians (anova). Hasil penelitian menunjukan bahwa penambahan susu skim dalam pengencer kuning telur memberikan pengaruh tidak nyata terhadap kualitas spermatozoa. Rata-rata motilitas spermatozoa tertinggi terdapat pada perlakuan P2, yaitu pengamatan 0 jam (87,8%), 12 jam (72,4%) dan 24 jam (58,2%). Rata-rata viabilitas spermatozoa tertinggi terdapat pada perlakuan P2, yaitu pengamatan 0 jam (78,8%), 12 jam (71%) dan 24 jam (53%). Peningkatan abnormalitas tertinggi didapatkan pada perlakuan P0 dengan rata-rata 0 jam (8,8%), 12 jam (12,2%) dan 24 jam (15,2%).Penambahan level susu skim dalam pengencer kuning telur mampu mempertahankan kualitas spermatozoa sapi Donggala selama 24 jam pada penyimpanan suhu 5oC. This study aimed to determine the effect of skim milk levels in egg yolk NaCl diluents on the cement quality of Donggala cows during storage at a temperature of 5oC. This research has been carried out at the Laboratory of the Technical Implementation Unit of the Livestock Breeding Service (UPTD), Central Sulawesi Province, in January-March 2022. Semen comes from 1 male Donggala cow with good reproductive conditions, aged 3 years with a body weight of 300 kg. The study design used a Complete Randomized Design (RAL) with 4 treatments: P0 = 95% physiological NaCl + 5% yolk; P1 = 88% physiological NaCl + 5% yolk + 7% skim milk; P2 = 87% physiological NaCl + 5% yolk + 8% skim milk; P3 = 86% physiological NaCl + 5% yolk + 9% skim milk. Each treatment is repeated 5 times. The observed variables are motility, viability, and abnormality. Data analysis with variance analysis (anova). The results showed that the addition of skim milk in the yolk thinner had an unreal influence on the quality of spermatozoa. The highest average spermatozoa motility was found in the P2 treatment, which was observed at 0 hours (87.8%), 12 hours (72.4%) and 24 hours (58.2%). The highest average viability of spermatozoa was found in the P2 treatment, namely observations of 0 hours (78.8%), 12 hours (71%) and 24 hours (53%). The highest increase in abnormalities was obtained in the P0 treatment with an average of 0 hours (8.8%), 12 hours (12.2%) and 24 hours (15.2%).
