Abstrak
ABSTRACT Background: Fixation is a crucial stage in cytopathology for diagnostic accuracy. 96% ethanol as a standard fixative is often limited in availability and cost. This study aimed to compare the effectiveness of 70%, 80%, and 96% alcohol in maintaining the cellular detail quality of pleural effusion preparations, in order to recommend an effective and affordable fixative alternative. Objective: To determine and statistically compare the effectiveness of the use of alcohol fixative 70%, 80%, and 96% on the detailed quality of cellular morphology preparations of pleural effusion cytopathology with Papanicolaou staining. Methods: This Post-Test Only quantitative laboratory experimental study used a single sample of pleural effusion divided into three dependent treatment slides. Data were analyzed using Cochran's Q Test. Results: 96% alcohol descriptively produced the highest good quality (67.3%) compared to 80% (50.0%) and 70% (48.1%). However, comparative analysis showed no statistically significant difference between the three concentrations (p = 0.074). Conclusions: The quality of the preparation's cellular morphological details did not differ significantly at alcohol concentrations of 70%, 80%, and 96% (p = 0.074). 70% and 80% alcohol are recommended as effective and reliable fixative alternatives when 96% alcohol is limited.
