ABSTRACT Introduction: Diabetes mellitus (DM) is a metabolic disease characterized by increased blood glucose levels (hyperglycemia), one of which is blood glucose 2 hours postprandial. Hyperglycemic blood glucose levels 2 hours postprandial are above 200 mg/dl, measured 2 hours after eating. To prevent complications of diabetes mellitus, it is necessary to give antidiabetic therapy. Garlic contains active chemical compounds in the form of allicin and flavonoids which can act as antidiabetics. Objective: The objective of this research is to find out the effect of garlic extract (Allium sativum Linn) on blood glucose levels 2 hours postprandial (GD2JPP) rats (Rattus norvegicus) in an alloxan-induced diabetic model. Methods: This research was true experimental research with a pre-post test control group design approach. 15 rats were adopted and divided into 5 groups: the negative control group was given 0.5% NaCMC, the positive control group was given metformin 45mg/kgBW and the garlic extract test group was 80 mg/KgBW, 200 mg/kgBW, and 400 mg/KgBW. Results: The results of the repeated ANOVA test show a significant difference in rats given the treatment with a value of p = 0.000 (p < 0> 0.05) and significantly different in the positive control group and garlic extract (p < 0.05). Conclusion: Based on the results above, the research concludes that a garlic extract dose of 400 mg/KgBW is the most significant dose in reducing blood glucose levels 2 hours post-prandial.
