Abstrak Background: Type 2 diabetes mellitus is a glucose metabolism disorder caused by insulin resistance. Insulin resistance triggers a metabolic syndrome with dyslipidemia, characterized by increased blood LDL levels. Moringa leaf (Moringa oleifera) can be used as an alternative treatment to decrease LDL levels because it has the bioactive compound quercetin. Purpose: This research aims to investigate the effectiveness of quercetin in moringa leaf and the optimum dosage of moringa leaf extract in decreasing blood LDL levels of rat models with type 2 diabetes mellitus. Methods: This research was conducted using a true experimental design, specifically a pretest-posttest control group design. The sample of this research was experimental animal models with type 2 diabetes mellitus that was established by using streptozotocin induction and a high-fat diet. Two-way analysis of variance (ANOVA) was conducted with Bonferroni post hoc tests to examine the differences between pre-test and post-test groups. Results: Administration of moringa leaf extract at a dosage of 400 mg/kg of body weight and 800 mg/kg of body weight decreased the average blood LDL levels of rat models with type 2 diabetes mellitus with a difference of 55,97 mg/dL and 69,315 mg/dL. The result of the Two-Way ANOVA test showed that the P-value was 0,0186 (p<0,05), indicating a significant difference in blood LDL levels before and after administering moringa leaf extract. The Bonferroni post hoc test showed that the group which received moringa leaf extract at a dosage of 800 mg/kg of body weight had a P-value of 0,0324 (p<0,05). This result showed the optimum dosage of moringa leaf extract in decreasing blood LDL levels. Conclusion: The quercetin in moringa leaf (Moringa oleifera) effectively decreases blood LDL levels of rat models with type 2 diabetes mellitus at the dosage of 800 mg/kg of body weight better than 400 mg/kg of body weight.
