Abstrak The skin of pomegranate fruit (Punica granatum L.) is known as a traditional medicine for stomachache because of intestinal worms bleeding, inflammation of the throat and gastric pain. The study aims to derermine total levels of flavonoid, total phenolic and potentional antioxidant pomegranate skin (Punica granatum L.) with variations in the method of extraction. Extraction skin of pomegranate fruit done with maceration and soxhletation using ethanol 96%, and infusion using distilled water. Assay of total flavonoids using method aluminum chloride with quersetin as a compare. Levels of total phenolic using the Folin-ciocalteau method with gallic acid as a compare. The antioxidant activity was determined by testing the captured of radical 2,2- diphenyl-1-picrylhydrazyl (DPPH). Levels of total flavonoids in skin extract of pomegranate fruit by maceration 23,69 QE/g, soxhletation 28,78333 QE/g, infuse 22,92666 QE/g, while the total phenolic in skin extracts of pomegranate fruit by maceration 22,68666 GAE/g, soxhletation 27,00666 GAE/g and infuse 23.42 GAE/g. The test results of antioxidant activity with DPPH method showed IC50 skin extract of pomegranate fruit by macerated 48,67 ?g/mL (a powerful antioxidant activity), soxhletation 49,28 mg/mL (a powerful antioxidant activity), and infuse 56,10 mg/mL (a powerful antioxidant activity).
