Abstrak ABSTRAK Limbah jerami jagung dan limbah biji kopi sebagai media dan sumber nutrisi yang dibutuhkan untuk pertumbuhan miselium jamur tiram putih (Pleurotus ostreatus). Penelitian ini telah dilaksanakan di Laboratorium Mikrobiologi Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Tadulako. Tujuan penelitian ini mengetahui pertumbuhan miselium serta karakteristik jamur tiram putih (Pleurotus ostreatus) pada formulasi media limbah jerami jagung dan limbah biji kopi. Penelitian ini didesain dalam Rancangan Acak Lengkap (RAL), terdiri dari tujuh perlakuan dan tiga kali ulangan yaitu P1 (serbuk jerami jagung 100%), P2 (serbuk jerami jagung 95% + serbuk limbah kopi 5%), P3 (serbuk jerami jagung 90% + serbuk limbah kopi 10%), P4 (serbuk jerami jagung 85% + serbuk limbah kopi 15%), P5 (serbuk jerami jagung 80% + serbuk limbah kopi 20%), P6 (serbuk jerami jagung 75 % + serbuk limbah kopi 25%), P7 (serbuk gergaji 70% + dedak padi 20% + tepung jagung 10% (kontrol positif)). Hasil penelitian menunjukkan laju pertumbuhan miselium jamur tiram putih perlakuan P3 memperlihatkan pertumbuhan yang sangat cepat dan perlakuan P6 menunjukkan pertumbuhan yang sangat lambat. Jumlah koloni yang tertinggi yaitu pada perlakuan P3 6,4x1011CFU/gr dan jumlah koloni yang terendah pada perlakuan P6 2,66x1011CFU/gr. Perlakuan P3 membutuhkan waktu yang lebih cepat untuk memenuhi botol kultur yakni 30 hari sedangkan P6 membutuhkan waktu lebih lama sampai memenuhi botol kultur yaitu 48 hari. Kata kunci: Jamur tiram putih, Jerami jagung, Limbah kopi, Media pertumbuhan ABSTRACT Corn straw waste and coffee bean waste as a medium and source of nutrients needed for the growth of mycelium white oyster mushroom (Pleurotus ostreatus). This research has been carried out at the Microbiology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Tadulako University. The purpose of this study was to determine the growth of mycelium and the characteristics of white oyster mushroom (Pleurotus ostreatus) in the formulation of corn straw and coffee bean waste media. This study was designed in a completely randomized design (CRD), consisting of seven treatments and three replications, namely P1 (100% corn straw powder), P2 (95% corn straw powder + 5% coffee waste powder), P3 (90 corn straw powder). % + 10% coffee waste powder), P4 (85% corn straw powder + 15% coffee waste powder), P5 80% corn straw powder + 20% coffee waste powder, P6 75% corn straw powder + coffee waste powder 25%), P7 (70% sawdust + 20% rice bran + 10% corn flour (positive control)). The results showed that the growth rate of white oyster mushroom mycelium in P3 treatment showed very fast growth and P6 treatment showed very slow growth. The highest number of colonies was in treatment P3 6,4x1011CFU/gr and the lowest number of colonies was in treatment P6 2,66x1011CFU/gr. Treatment P3 takes a shorter time to fill the culture bottles, which is 30 days, while P6 takes a longer time to fill the culture bottles, which is 48 days.
