Abstrak
Free radicals were one of the main causes of oxidative stress, capable of damaging cells and triggering degenerative diseases. The body possesses endogenous antioxidant systems such as superoxide dismutase (SOD) and glutathione stimulation hormone (GSH) to neutralize free radicals. This study aimed to analyze the effect of nanocapsulated ethanolic extract of papaya leaves (Carica papaya L.) on SOD and GSH levels in benzo[?]pyrene induced white rats (Rattus norvegicus). The extract was formulated into nanocapsules using kappa-carrageenan at ratios of 1:1, 1:2, and 2:1. Characterization was performed using PSA and FTIR, and in vivo testing was conducted to assess antioxidant effects. Particle sizes ranged from 228.7 to 340.2 nm. SOD and GSH levels were measured using the ELISA Reader method. The 1:2 ratio showed the highest SOD (3.348 ng/mL) and GSH (10.018 ng/mL), followed by 1:1 (SOD 2.801 ng/mL; GSH 7.914 ng/mL) and 2:1 (SOD 2.512 ng/mL; GSH 7.361 ng/mL). The 1:2 formulation was the most effective in enhancing endogenous antioxidants. Keywords: Nanocapsule, Papaya leaf, SOD, GSH, Oxidative stress, Benzo[?]pyrene
