Abstrak
Nur Ainun Fitri, E 281 17 119. Induction of Red Paprika (Capsicum annum. L) Callus from Hypocotyls in Various Combinations and Concentrations of Growth Regulators. (Supervised by; Zainuddin Basri and Maemunah, 2023). Plant growth in tissue culture is determined by various factors, such as the type of explant and the composition of the ZPT used. This study aims to examine the effect of the combination and concentration of growth regulators on callus induction in red peppers from the hypocotyl. This research was carried out at the Biotechnology Laboratory, Faculty of Agriculture, Tadulako University from October to December 2023. Using a Single Factor Completely Randomized Design with the type of explant used, namely hypocotyl. The composition of growth regulators consists of six combinations and concentrations, namely 5 ppm Bap + 0.25 ppm IAA; 5 ppm BAP + 0.5 ppm IAA; 5 ppm TDZ + 0.5 ppm GA3; 5 ppm TDZ + 1 ppm GA3; 7.5 ppm Kinetin + 0.5 ppm GA3; 7.5 ppm Kinetin + 1 ppm GA3. Each treatment combination was repeated three times so that there were 18 experimental units, each experimental unit was planted with six explants so there were 108 explants. The variables observed were when callus appeared, the percentage of explants forming callus, callus texture, callus color. The data were analyzed using diversity analysis and the results of the diversity analysis showed real or very real effects followed by conducting an Honest Significant Difference test at the 5% level. The results of the research can be concluded that the combination and concentration of growth regulators which are more effective for callus induction in red peppers from hypocotyl explants has a very good effect. Giving 5 ppm TDZ + 1 ppm GA3 (M4) resulted in the fastest callus emergence, namely 15.67 days. Giving 5 ppm TDZ + 1 ppm GA3 (M4) resulted in a percentage of explants forming callus, namely 100%. Administration of 5 ppm TDZ + 1 ppm GA3 (M4) forms a compact callus texture with a yellow callus color.
