Restu Dwi Nugroho, (E 202 21 032), Identification of Local Upland Rice of Central Sulawesi Using PCR Techniques and Chemical Properties. (Supervised by Rostiati, Nur Edy) Upland rice has long been cultivated by farming communities in Central Sulawesi, but in the food crop agricultural development program upland rice has received less attention so that it has an impact on the lack of development of upland rice cultivation which is reflected in the low production of upland rice in Central Sulawesi, which in 2022 is approximately 13,420 hectares with a production target of 39,328 tons, and productivity of 29.31 quintals per hectare. This target increased compared to the realization of production in 2021 which reached 37,814 tons with a productivity of 37.87 quintals per hectare from a harvest area of 9,985 hectares. Efforts to encourage the development of upland rice in Central Sulawesi are to optimize planting materials or superior upland rice seeds. Therefore, one approach to identify the diversity of upland rice in Central Sulawesi is to identify the genetic diversity and chemical properties of local upland rice cultivars in Central Sulawesi. The purpose of this research is to identify the chemical properties of Central Sulawesi upland rice cultivars and identify genetic diversity in Central Sulawesi upland rice cultivars. This research is a type of experimental research with quantitative descriptive approach. Quantitative descriptive analysis was conducted through direct experiments on the object of research, namely 5 cultivars of upland rice of Central Sulawesi, namely Paebohe, Menso, Kalendeng, Pulu Konta, and Puyutas cultivars to determine chemical properties and genetic identification through PCR method with forward primer rbcLa-F with the arrangement 5'-ATGTCACCACAA ACAGAAAC-3'. There are differences in the chemical content of each local upland rice cultivar of Central Sulawesi including the highest water content found in paebohe cultivar 9.88%, the highest ash and fat content in Kutivar Pulu konta which is 1.07% and 1.18?t content, the highest carbohydrate content in Puyutas cultivar 82.53%, the highest protein and amylose content in kalendeng cultivar respectively 10.34% and amylose 19.96% while menso cultivar has the highest amylopectin content which is 45.42 mg/ml. The farthest genetic distance is found in the Menso and Kalendeng cultivars which is 0.645 even though they are in one province. Puyutas, Pue konta and Kalendeng varieties are grouped into one node with a genetic distance of 0.289 and 0.500 and are still related to Paebohe cultivars with a genetic distance of 0.577.
